چکیده

Over the last decade our understanding of the role of ROS has progressed from the classical view of adverse toxic metabolic byproducts inadvertently associated with aerobic life to include the newly emerging role of signaling molecules regulating growth, development and coordinating responses to abiotic and biotic stress. A recent series of discoveries have given scientists new insights into ROS-dependent gene activation and the molecular mechanisms involved. The majority of information of the regulatory role of ROS on gene expression derived from experiments using: i) transgenic plants overexpressing or suppressing antioxidant genes in order to reduce or increase the intracellular ROS levels, respectively; ii) mutants impaired in ROS generation or scavenging; iii) direct application of ROS; iv) application of ROS generating compounds. Results of these experiments provided signifi cant information on ROS-dependent signaling pathways and ROS-responsive genes. A number of genes involved in defense, signal transduction, transcription, metabolism as well as cell structure have been identifi ed revealing a highly dynamic and redundant network of ROS-producing and ROS-scavenging genes. Antioxidant genes are central players in this network and their function has profound effect in controlling ROS levels and cellular redox balance. This chapter focuses on recent developments on the role of ROS in activation of gene expression with an emphasis on ROS-scavenging enzyme systems.

Rhizophagus irregularis regulates antioxidant activity and gene expression under cadmium toxicity in Medicago sativa

Cadmium (Cd) is a phytotoxic heavy metal (HM) that can induce generation of reactive oxygen species (ROS). Arbuscular mycorrhizal fungi (AMF) are considered as bio-ameliorators that help to mitigate HM-derived oxidative stress. The objective of this study was to assess AM fungus Rhizophagus irregularis on changes in enzymatic activity and transcription of antioxidants of Medicago sativa to Cd s...

Antioxidant gene responses to ROS-generating xenobiotics in developing and germinated scutella of maize.

There is circumstantial evidence implicating reactive oxygen species (ROS) in the highly ordered temporal and spatial regulation of expression of the Cat and Sod antioxidant genes during seed development and germination in maize. In order to understand and provide experimental data for the regulatory role of ROS, the expression patterns of the Cat1, Cat2, Cat3, GstI, Sod3, Sod4, and Sod4A genes...

Effects of Exercise Training and Consumption of Fenugreek Seed Extract on Expression of Oxidised LDL and ROS-related Genes in Patients with Coronary Artery Occlusion

Background and objectives: Sedentary life style and high-fat diet are one of the key causes of coronary artery disease. The aim of this study was to evaluate effects of periodic exercise and consumption of fenugreek seed extract on the expression of oxidized low-density lipoprotein (oxLDL) and reactive oxygen species (ROS) genes in patients with coronary artery occlusion. Methods: The present ...

Human Leukemic Cells performing Oxidative Phosphorylation (OXPHOS) Generate an Antioxidant Response Independently of Reactive Oxygen species (ROS) Production

Tumor cell metabolism is altered during leukemogenesis. Cells performing oxidative phosphorylation (OXPHOS) generate reactive oxygen species (ROS) through mitochondrial activity. To limit the deleterious effects of excess ROS, certain gene promoters contain antioxidant response elements (ARE), e.g. the genes NQO-1 and HO-1. ROS induces conformational changes in KEAP1 and releases NRF2, which ac...

Repression of sestrin family genes contributes to oncogenic Ras-induced reactive oxygen species up-regulation and genetic instability.

Oncogenic mutations within RAS genes and inactivation of p53 are the most common events in cancer. Earlier, we reported that activated Ras contributes to chromosome instability, especially in p53-deficient cells. Here we show that an increase in intracellular reactive oxygen species (ROS) and oxidative DNA damage represents a major mechanism of Ras-induced mutagenesis. Introduction of oncogenic...
